چکیده

Introduction Therapeutic cell treatments have been applied to a large number of pathological diseases. Tracking the engraftment of cells is key to planning and assessing cell therapies. Migration dynamics and tissue integration are commonly assessed by locating iron-oxide labeled cells with T2* weighted MRI. Interpretation of the resulting signal voids can be challenging in preclinical scanners, especially in regions of magnetic field inhomogeneities or susceptibility. Although techniques exist to obtain positive contrast of iron-oxide labeled cells, such as Off-Resonance Imaging [1] and Inversion-Recovery with ON-Resonant Water Suppression [2], their sensitivity to magnetic field inhomogeneities deter application at high B0. We present here a multi-echo technique that exploits both the T1 and T2-relaxation effect of iron to achieve positive contrast of SPIO-labeled cells at 9.4T.

Positive Contrast Imaging of Iron-Oxide Labeled Human Embryonic Stem (hES) Cell and Fibroblast using SWEET

Introduction: The MRI detection of superparamagnetic iron-oxide particles (SPIO) -labeled cells is important in cell-replacement therapies, which can help monitoring the migration/proliferation of cells following the transplantation. Cells loaded with SPIOs cause signal reduction (negative contrast) associated with dephasing due to the magnetic field inhomogeneity near the cells, therefore the ...

MR tracking of transplanted cells with "positive contrast" using manganese oxide nanoparticles.

Rat glioma cells were labeled using electroporation with either manganese oxide (MnO) or superparamagnetic iron oxide (SPIO) nanoparticles. The viability and proliferation of SPIO-labeled cells (1.9 mg Fe/ml) or cells electroporated with a low dose of MnO (100 microg Mn/ml) was not significantly different from unlabeled cells; a higher MnO dose (785 microg Mn/ml) was found to be toxic. The cell...

Mesenchymal stem cell labeling and in vitro MR characterization at 1.5 T of new SPIO contrast agent: Molday ION Rhodamine-B™.

In vivo detection of transplanted stem cells is requisite for improving stem cell-based treatments by developing a thorough understanding of their therapeutic mechanisms. MRI tracking of magnetically labeled cells is non-invasive and is suitable for longitudinal studies. Molday ION Rhodamine-B™ (MIRB) is a new superparamagnetic iron oxide (SPIO) contrast agent specifically formulated for cell l...

In vivo MRI using positive-contrast techniques in detection of cells labeled with superparamagnetic iron oxide nanoparticles.

Positive-contrast techniques are being developed to increase the detection of magnetically labeled cells in tissues. We evaluated a post-processing positive-contrast technique, susceptibility-gradient mapping (SGM), and compared this approach with two pulse sequences, a gradient-compensation-based "White Marker" technique and an off-resonance-based approach, inversion recovery on-resonance wate...

Application of Ultra-Short Echo Time Imaging for Visualization of SPIO-loaded Tumor Cells in Brain

Introduction: There has been recent interest in positive-contrast MRI methods for tracking cells labeled with superparamagnetic iron oxide (SPIO) nanoparticles. Due to signal variations caused by SPIO-induced field inhomogeneities, in tissues with highly concentrated iron labeled cells, T2* values can decrease to values below 1 millisecond. Therefore, the signal may have decayed to the noise le...
